Berberine Enhances Osseointegration in  Diabetic Rats

小檗碱对糖尿病大鼠种植体骨整合的影响

Abstract:

Objective: To study the effects of berberine on osseointegration of dental implants in type II diabetic rats. Method: Seven-month-old  (n=35)  male healthy SD rats were used in this study. 10 rats were randomly selected and assigned to healthy group (H group). In the remaining 25 rats, in order to construct the model of type II diabetes, high glucose diet and low dose of streptozotocin were provided, and finally, the model was successfully  established in 20 rats. 60 pieces of pure titanium implants were installed into bilateral femoral metaphysic of the diabetic rats and then divided into 2 groups: berberine treatment group (B group), no treatment group(N group). The body weight and fasting blood glucose were measured at 7-, 31-, 38-, 66- and 101- day respectively. The rats were sacrificed at 10 weeks after implantation. Micro-CT scan, VG staining and implant pull-out test were performed.VG staining of hard tissue slices, bone volume fraction (BV/TV), Tb.Th, trabecular number (Tb.N Tb.Sp), trabecular bone volume (TBV), bone-implant contact (BIC) and the maximum pullout force were measured for comparison. Results: ① The weight of rats in H Group increased in a time dependent manner. Rats in B group and N group, weight reached the maximum value at 31 days, then decreased gradually in B group, and decreased sharply in N group (P<0.05). ②Blood glucose: H group <B group <N group (P<0.05). ③BV/TV, Tb.Th, Tb.N, TBV, BIC and maximum pullout force: H group >B group >N group. Tb.Sp: H group <B group <N group  (P<0.05). Conclusion: Berberine can effectively reduce blood glucose level in type 2 diabetic rats and promotes osseointegration of dental implants.

摘要:

目的:研究小檗碱对Ⅱ型糖尿病大鼠种植体骨整合的影响。方法:35只7月龄SPF级雄性健康SD大鼠,随机抽取10只,分入健康组(H组)。剩余25只通过高糖饮食+小剂量链脲佐菌素(STZ)诱导,构建Ⅱ型糖尿病大鼠模型共计20只(5只失败),随机分为小檗碱治疗组(B组)和非治疗组(N组)。将60枚纯钛种植体植入大鼠双侧股骨干骺端,分别于第7、31、38、66、101天测量大鼠的体质量和空腹血糖值。种植术后10 周将大鼠处死,取股骨行Micro?鄄CT扫描、VG染色以及种植体拔出实验。观察VG染色硬组织切片,测量骨体积分数(BV/TV)、骨小梁厚度(Tb.Th)、骨小梁数目(Tb.N)、骨小梁间隙(Tb.Sp)、骨小梁容积率(TBV)、骨接触率(BIC)及最大拔出力。结果:①H组体质量随时间持续增长;B组和N组体质量31 d时达到最大值,此后B组体质量逐渐降低,N组体质量快速降低(P<0.05);②血糖值高低比较:H组<B组<N组(P<0.05)。③BV/TV、Tb.Th、Tb.N、TBV、BIC及最大拔出力比较,均为H组>B组>N组(P<0.05);Tb.Sp值H组<B组<N组(P<0.05)。结论:小檗碱可有效降低Ⅱ型糖尿病大鼠血糖,并促进种植体的骨整合。
