Abstract: Objective: The aim of this study was to observe the expression changes of tumor necrosis factor α (TNFα) and fibroblast growth factor 2 (FGF2) in rat submandibular glands(rSMG) after irradiation. Methods: 19 male wistar rats  were randomly divided into three groups: group A is no IR group (control group), group B is IR 3-day group, and group C is IR 30-day group. The rats submandibular area were radiated with a single dose of 15 Gy. The animals were anesthetized and sacrificed. rSMG were removed and processed by immunohistochemistry for the detection of TNFα and FGF2. Results: As compared to the control group, TNFα and FGF2 expression were more intensive in IR 3-day group, and most intensive in IR 30-day group. Conclusion: Irradiation induced an increase of the expression of TNFα and FGF2 in rat submandibular glands.

摘要: 目的:观察照射后大鼠颌下腺TNFα和FGF2的表达变化,探讨放疗后唾液腺纤维化的可能分子机制。方法:19只大鼠随机分为对照组(只麻醉不照射)、照射组。照射组用15 Gy的X射线进行颌下腺区局部照射1次,照射后3 d和30 d处死大鼠取颌下腺,4%多聚甲醛固定、石蜡包埋、切片,免疫组化染色观察颌下腺TNFα和FGF2表达的变化。结果:照射后3 d和30 d大鼠颌下腺TNFα和FGF2表达均升高,30 d组高于3 d组。结论:照射后唾液腺纤维化可能与TNFα和FGF2表达升高有关。
