长链非编码RNA HOTAIR在口腔鳞癌组织中的表达及临床意义

Abstract: Objective: To investigate the expression of long non-coding RNA HOTAIR in oral squamous cell carcinoma (OSCC) and its clinical significance. Methods: 71 patients with OSCC were selected. Real-time fluorescence quantitative PCR was used to detect the expressions of HOTAIR mRNA in OSCC and normal oral mucosal tissues. The patients were followed up after surgery, the deadline time was March 31, 2017. Survival analysis was performed using Kaplan-Meier method and Log-rank test. Multi-factor analysis was performed using Cox risk proportional regression model. Results: The relative expression level of HOTAIR mRNA in OSCC tissues was 2.36±0.19, while in the control group was 1.53±0.12, the difference was statistically significant (t=30.758, P=0.000). The relative expression level of HOTAIR mRNA in OSCC tissues was correlated with clinical stage, degree of differentiation, TNM stage and local recurrence (P<0.05). The 5-year survival rate in the low expression group was 42.11%, the median survival time was 49.0 months, while in the high expression group were 18.75% and 25.90 months, respectively. The Log-Rank test showed the difference was statistically significant ( χ2=4.871, P=0.027). Cox multivariate analysis showed that clinical stage, lymph node metastasis, local recurrence and HOTAIR mRNA expression were independent prognostic factors in patients with OSCC. Conclusion: The HOTAIR was highly expressed in OSCC tissues. It was closely related to tumorigenesis and progression, and was an independent factor affecting the prognosis of patients.

摘要: 目的:探讨长链非编码RNA HOTAIR在口腔鳞癌(OSCC)组织中的表达及临床意义。方法:选取2010-02—2012-03期间,在我院就诊的OSCC患者71例,利用实时荧光定量PCR技术检测OSCC组和正常口腔黏膜组织中HOTAIR mRNA表达,术后对患者随访,截止时间为2017-03-31。生存分析采用Kaplan-Meier法和Log-rank检验,多因素分析采用Cox风险比例回归模型。结果:OSCC组织中HOTAIR mRNA相对表达量为2.36±0.19,对照组结果为1.53±0.12,两组比较,差异有统计学意义(t=30.758,P=0.000);OSCC组织中HOTAIR mRNA相对表达量与临床分期、分化程度、TNM分期、局部复发有关(P<0.05);低表达组患者5年生存率42.11%,中位生存时间49个月,高表达组患者5年生存率18.75%,中位生存时间25.90个月,Log-Rank检验差异有统计学意义(χ2=4.871,P=0.027)。Cox多因素分析结果显示,临床分期、淋巴结转移、局部复发和HOTAIR mRNA表达量是影响OSCC患者预后的独立因素(P<0.05)。结论:OSCC组织中HOTAIR呈高表达,与肿瘤发生、进展密切相关,是影响患者预后的独立因素。
