Abstract: Objective: To explore the effects of concentrated growth factor(CGF)on the healing of gingival soft tissue, as well as the underlying cellular biological mechanism defects. Methods: About 1.0 cm×1.0 cm gingival soft tissue wound was made in the healthy female mini-pig from the top of the mandibular alveolar crest to the labial vestibule groove. The wound was covered with CGF membrane (experimental group) or gelatin sponge (control group). The healing of the wounds in each group was observed after surgery. Gingival soft tissues were taken for histological observation on the 3rd, 7th, 14th, and 28th days. The gingival fibroblasts were isolated from the gingival soft tissue of twelve-weeks and male SD rats for experimental or control group. The proliferation and migration were compared with CCK-8 or cell wound-healing test, respectively. Results: The edema in the marginal experimental group was lower than that in the control group, accompanied by significantly less infiltration of inflammatory cells in the connective tissues. Consistently, the experimental group showed the better and more rapid wound healing than that in the control group. The epithelium of the experimental groups indicated orthokeratinization, however the controls indicated parakeratinized epithelia. The increased migration of fibroblasts, as induced by CGF in vitro, may facilitated the healing rate of experimental group as well. Conclusion: CGF can promote wounds healing of gingival soft tissue and reduce inflammation. CGF extract can promote the proliferation and migration of gingival fibroblasts.

摘要: 目的:研究浓缩生长因子(concentrated growth factor,CGF)在促进牙龈软组织缺损修复过程中的作用及其细胞生物学机制。方法:在健康雌性小型猪的下颌牙槽嵴顶至唇侧前庭沟制作一面积约1.0 cm×1.0 cm深达骨膜的牙龈软组织缺损。于缺损处覆盖CGF膜(实验组)或明胶海绵(对照组)。术后观察各组缺损的愈合情况,并于术后第3、7、14、28 d取术区牙龈软组织进行组织学观察。取健康雄性12周大鼠牙龈分离培养并鉴定牙龈成纤维细胞,CCK-8及划痕实验观察CGF提取液对其增殖迁移的影响。结果:实验组术区边缘红肿等炎症反应程度比对照组程度轻,愈合更好。术后组织学观察见实验组创伤区炎症细胞浸润明显少于对照组,愈合速度较对照组明显加快。实验组上皮表面以正角化为主,对照组以不全角化为主。CGF提取液处理组大鼠的牙龈成纤维细胞,增殖及划痕愈合速度均比对照组加快。结论:CGF可以减轻炎症反应并促进牙龈软组织缺损修复。CGF提取液可以促进牙龈成纤维细胞的增殖迁移。
