TLR4与LPS诱导的大鼠口腔黏膜分泌TNF-α的相关性研究

摘要: 目的:观察Toll样受体-4(Toll like receptors 4,TLR4)与脂多糖(lipopolysaccharide,LPS)诱导的,大鼠口腔黏膜上皮细胞分泌肿瘤坏死因子-α(tumor necrosis factor-α, TNF-α)的相关性。方法:选取SPF级SD大鼠15只,根据随机数字表法将其随机均分为5组,分别为采用不同浓度和不同时间的LPS刺激大鼠口腔黏膜,检测大鼠口腔黏膜上皮细胞培养上清液中TNF-α的表达情况,检测细胞内TLR4、MyD 88的表达变化。结果:经过LPS刺激后,TNF-α蛋白质的表达量随着LPS刺激浓度的增加而逐渐升高,呈现出明显的剂量依赖性效应。另外,二者与LPS刺激的持续时间呈现出一定的时间依赖性,TNF-α蛋白质的表达量在12 h达到峰值。LPS能够明显增加细胞内TLR4和MyD88蛋白质的表达。结论:LPS诱导并促进大鼠黏膜上皮细胞分泌TNF-α的情况,可能与TLP4调控有关。

Abstract: Objective: To investigate the effect of Toll-like receptors 4 (TLR4) on the secretion of tumor necrosis factor-α (TNF-α) in oral epithelial cells induced by lipopolysaccharide (LPS) Α and its mechanism. Methods: Fifteen SD rats were randomly divided into five groups according to the random number table method. The oral mucosa was stimulated by LPS at different concentrations and different time length.  The expression of TLR4 and MyD88 in the oral epithelian cells and expression of TNF-α within the supernatant were detected by immunohistochemistry. Results: After LPS stimulation, the expression of TNF-α protein increased gradually correlated with the increase of LPS stimulation concentration, showing a dose-dependent manner. In addition, they also showed a time-dependent manner in certain degree.  TNF-α protein expression peaked at 12 h. LPS could significantly increase the expression of intracellular TLR4 and MyD88 proteins. Conclusion: LPS induced and promoted TNF-α secretion in rat mucosal epithelial cells may be related to TLP4 regulation.
