NLRP3和Caspase-1在颞下颌关节骨关节炎滑膜中的表达

Expression of NLRP3 and Caspase-1 in synovium of temporomandibular joint osteoarthritis

摘要:

目的:探究NLRP3炎症小体(NACHT, LRR and PYD domains-containing protein 3 inflammasome)和胱天蛋白酶-1(cysteinyl aspartate specific proteinase,Caspase-1)在颞下颌关节骨关节炎(temporomandibular joint osteoarthritis,TMJOA)患者滑膜细胞中的表达。方法:分离人颞下颌关节滑膜组织并进行滑膜细胞培养,另取3例髁突肥大患者的滑膜组织作为对照组。实时定量聚合酶链式反应(real-time quantitative polymerase chain reaction,RT-PCR)法检测滑膜细胞中NLRP3和Caspase-1的基因表达,Western blot检测滑膜细胞中NLRP3和Caspase-1的表达。结果:TMJOA患者滑膜细胞中的NLRP3和Caspase-1的表达显著高于对照组(P<0.05)。结论:NLRP3和Caspase-1在TMJOA滑膜细胞中高表达,说明它们与TMJOA发生发展密切相关,这将有助于进一步阐明其分子机制。

Abstract:

Objective: To investigate the expression of NLRP3(NACHT, LRR and PYD domains-containing protein 3 inflammasome) and Caspase-1 in the synovium of temporomandibular joint osteoarthritis(TMJOA). Methods: Synovial tissues were collected from TMJ of patients and synovial cells were cultured. Synovial tissues of three patients with condylar hypertrophy were taken as control group. Real-time PCR was used to examine the gene expression of NLRP3 and Caspase-1 in synovial cells. The protein expressions of NLRP3 and Caspase-1 in synovial cells were detected by Western blot. Results: The expressions of NLRP3 and Caspase-1 in the synovial cells of TMJOA were significantly increased than the control group(P<0.05). Conclusion: NLRP3 and Caspase-1 were increased in the synovial cells of TMJOA and closely related to the occurrence and development of TMJOA, which will help to further clarify its molecular mechanism.
