miR-429对头颈部鳞状细胞癌增殖的影响

miR-429 Inhibits Cell Growth of Head and Neck Squamous Cell Carcinoma

摘要: 目的:研究microRNA-429(miR-429)对人头颈部鳞癌细胞增殖的影响。方法:通过qRT-PCR分析19例头颈部鳞状细胞癌患者肿瘤组织与癌旁正常组织标本中miR-429的表达。通过miR-429 mimics 或miR-429 ASO上调或抑制人喉癌细胞株(Hep-2)和人舌鳞状细胞癌株(SCC-9)细胞中miR-429的水平。用MTT实验分析上调或抑制miR-429的表达水平对Hep-2和SCC-9细胞增殖的影响,并利用生物信息学方法预测miR-429的靶基因。结果:qRT-PCR结果表明头颈部鳞状细胞癌组织中miR-429表达水平较低;上调miR-429表达能抑制Hep-2和SCC-9细胞的增殖,而抑制miR-429表达能促进Hep-2和SCC-9细胞的增殖。生物信息学方法预测miR-429的靶基因为ZEB1基因。结论: miR-429可能通过ZEB1基因抑制头颈部鳞状细胞癌的增殖。

Abstract: Objective: To study whether the effect of miR-429 was related to the proliferation of head and neck squamous cell carcinoma (HNSCC) cells. Method: The levels of miR-429 in 19 HNSCC biopsied tissues and the matched adjacent normal tissues were analyzed by qRT-PCR. The miR-429 levels in Hep-2 or SCC-9 were over-expressed by miR-429 mimics or miR-429 ASO transfection. The cellular proliferation was analyzed by MTT assay. The targeted genes of miR-429 were predicted by bioinformatics algorithms. Results: The level of miR-429 in HNSCC tissues was lower than the matched normal tissue. Overexpression of miR-429 decreased the proliferation of Hep-2 and SCC-9 cells, and inhibition of miR-429 levels promoted the proliferation of Hep-2 and SCC-9 cells in a time-dependent manner. ZEB1 is likely to be the targeted gene of miR-429 in HNSCC.  Conclusion: miR-429 may inhibit HNSCC proliferation via ZEB1.
