姜黄素对口腔鳞癌SCC-25细胞EGFR信号通路表达的影响

Curcumin Inhibits Human Oral Cancer Cell Growth by Suppressing EGFR Phosphorylation

摘要: 目的:探讨姜黄素对口腔鳞癌SCC-25细胞EGFR信号通路表达的影响及其相关的分子机制。方法:不同浓度姜黄素作用细胞24 h,MTT法检测姜黄素对细胞增殖作用的影响,Transwell实验检测EGF 和姜黄素共同作用下细胞侵袭能力的变化,Western blot检测姜黄素对SCC-25细胞EGFR活性及下游信号分子(Akt、ERK1/2 和 STAT3 )表达的影响。结果:不同浓度姜黄素作用SCC-25细胞24 h后,均可抑制SCC-25细胞增殖,亦可以抑制EGF诱导的细胞侵袭转移。Western blot结果显示姜黄素对SCC-25细胞总EGFR表达无影响,但可以剂量依赖性抑制SCC-25细胞p-EGFR表达,同时可以剂量依赖性抑制EGFR信号通路下游转录因子Akt、ERK1/2 和 STAT3磷酸化。结论:姜黄素可剂量依赖性抑制SCC-25细胞EGFR磷酸化活性及其下游信号通路分子(Akt、ERK1/2、STAT3)的磷酸化。

Abstract: Objective: To study whether Curcumin was associated with the signaling pathway expression of EGFR of human oral squamous cell carcinoma (OSCC) SCC-25 cells and its molecular mechanisms. Methods: The efficacy of curcumin on proliferation in SCC-25 cell line were investigated. The inhibition effect of curcumin on EGF-induced SCC-25 cells invasion were explored by transwell assay. The efficacy of curcumin on the activition of EGFR and its downstream signaling molecules Akt, ERK1/2 and STAT3 were investigated by western blot. Results: Curcumin inhibited SCC-25 cells proliferation and EGF-triggered SCC-25 cells invasion. Curcumin regulated the p-EGFR and EGFR downstream signaling molecules including Akt, ERK1/2 and STAT3. Conclusion: Curcumin reduced SCC-25 cells proliferation and invasion through inhibiting the phosphorylation of EGFR and EGFR downstream signaling molecules Akt, ERK1/2 and STAT3.
