BDNF/TrkB/Akt/p-RPS6信号通路在口腔上皮牙源性病变患儿中的作用及免疫反应分析

Analysis of BDNF/TrkB/Akt/p-RPS6 signaling pathway in children with oral epithelial odontogenic lesion

摘要: 目的: 分析BDNF/TrkB/Akt/p-RPS6信号通路在儿童口腔上皮牙源性病变中的作用及免疫反应。方法: 以2018年6月—2020年6月间本院收治的50例口腔上皮牙源性病变患儿的口腔上皮病变组织标本(病例组)和50例健康儿童的口腔上皮组织标本(对照组)为研究对象,均行BDNF/TrkB/Akt/p-RPS6信号通路蛋白检测、免疫功能T细胞亚群(CD3+、CD4+、CD4+/CD8+)检测和免疫球蛋白指标(IgG、IgA、IgM)检测,对比对照组和病例组上述指标的差异,并采用Pearson相关性分析法分析BDNF/TrkB/Akt/p-RPS6信号通路蛋白与免疫功能相关指标的相关性。结果: 病例组的脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)、酪氨酸激酶受体B(tyrosine kinase receptor B,TrkB)、蛋白激酶B(protein kinase B,PkB,又称Akt)、磷酸化S6核糖体蛋白抗体(anti-phospho-S6 ribosoma protein,p-RPS6)的平均光密度值均高于对照组(P<0.05)。病例组的免疫功能T亚群细胞(CD3+、CD4+、CD4+/CD8+)、免疫球蛋白指标(IgG、IgA、IgM)数据值均低于对照组(P<0.05)。BDNF、TrkB、Akt、P-RPS6的平均光密度值与免疫功能T细胞亚群(CD3+、CD4+、CD4+/CD8+)数据值、免疫球蛋白指标(IgG、IgA、IgM)数据值呈负相关(均P<0.05)。结论: 口腔上皮牙源性病变患儿存在明显的BDNF/TrkB/Akt/p-RPS6信号通路相关蛋白表达异常和免疫功能抑制,且免疫功能抑制与BDNF/TrkB/Akt/p-RPS6信号通路蛋白异常表达相关。

Abstract:

Objective: To analyze the role and immune response of BDNF/TrkB/Akt/p-RPS6 signaling pathways in children′s oral epithelial odontogenic lesions. Methods: Tissue specimens from 50 children with oral epithelial odontogenic lesions (odontogenic lesion group) and 50 oral epithelial tissue samples (control group) who were treated in our hospital from June 2018 to June 2020 were selected. In all samples, proteins in BDNF/TrkB/Akt/p-RPS6 signaling pathway, immune function T cell subsets(CD3+, CD4+, CD4+/CD8+) and immunoglobulin indexes (IgG, IgA, IgM) were detected to compare the differences of the above indexes between healthy group and odontogenic lesion group. Pearson correlation analysis was used to analyze the correlation between signal pathway protein and immune function related indexes. Results: The average optical density of brain-derived neurotrophic factor (BDNF), tyrosine kinase receptor (TrkB),protein kinase B (PkB, also know as Akt) and anti-phospho-S6 ribosoma protein(p-RPS6) in odontogenic lesion group was higher than that in control group (P<0.05). The data of immune function T cell subsets(CD3+, CD4+, CD4+/CD8+) in odontogenic lesion group were lower than those in control group(P<0.05). The data of immunoglobulin indexes (IgG, IgA, IgM) in odontogenic lesion group were lower than those in control group(P<0.05). The mean optical density value of BDNF, TrkB, Akt and p-RPS6 was negatively associated with the data of immune functional T cell subsets(CD3, CD4, CD4/CD8) and immunoglobulin indexes(IgG, IgA, IgM)(all P<0.05). Conclusion: There were obviously abnormal expression of BDNF/TrkB/Akt/p-RPS6 signal pathway related proteins and immunosuppression in patients with oral epithelial odontogenic lesions, and immunosuppression was related to abnormal expression of BDNF/TrkB/Akt/p-RPS6 signal pathway proteins.
