磨牙咬合升高对小鼠短期内焦虑情绪的影响

Occlusion Vertical Rising of Molars Increased Anxiety Temporally in Mice

摘要: 目的:建立磨牙咬合升高小鼠模型,观察其短期内焦虑样情绪及臂旁核、杏仁核中神经元激活水平的变化。方法:8周龄C57BL/6J雄性小鼠,随机分为2组:假手术组和咬合升高组。造模后连续记录体重及进食量;于第1天、第3天、第7天及第14天,行高架十字迷宫测试、旷场测试、新环境抑制进食测试、社交探索测试等焦虑样情绪相关的行为学检测;免疫荧光染色观察小鼠杏仁核中央区Fos蛋白表达水平。结果:磨牙区咬合升高对小鼠体重、进食量及基础活动能力并无短期影响。在处理后7、14 d,咬合升高组小鼠焦虑样行为明显增加。与假手术组相比,咬合升高导致小鼠臂旁核、杏仁核神经元激活水平升高。结论:磨牙区咬合升高会导致小鼠短期内焦虑样情绪水平增加。

Abstract: Objective: To explore the onset and development of affective disorders concomitant with occlusion vertical rising (OVR) in mice models. Methods: 8-week-old C57BL/6J male mice were randomly assigned into two groups: OVR group and sham group. On postoperative Day1/Day3/Day7/Day14, the anxiogenic phenotypes were verified via animal behavior during the elevated plus maze test, and by additional analysis of the open field test, the novelty-suppressed feeding test and the social exploration test. The expression of Fos in central amygdaloid nucleus (CeA) and parabrachial nucleus (PBN) were detected by immunofluorescence. Results: Over perioperative period, the weight, food intake and locomotor activity in two groups did not show significant difference. Occlusion vertical rising group exhibited more anxiety-like behaviors compared to sham group in multiple tests. Meanwhile, the level of Fos-positive cells in CeA and PBN in occlusion vertical rising group were higher than that in sham group. Conclusion: The results indicate that occlusion vertical rising may enhance to more anxiety-related behaviors in mice temporally.
