The Mechanism of ERK and Integrinβ1 Mediated Morphological Changes in Mandibular Condyle after Long-term Mandibular Functional Protrusion in Rats

生长期大鼠下颌功能前伸后髁突的骨形态学改变及其ERK与整合素β1表达的变化

Abstract: Objective: To establish a mandibular protrusion animal model in SD rats and investigate the changes of ERK and integrin β1, then to analyze the morphological changes of mandibular condyles in the mandibular growth stage. Methods: 42 SPF healthy male SD rats, 4 week old, were divided into control group and experimental group randomly, 3 rats in each group, and there are 14 groups. The experimental period were 1, 3, 7, 14, 21, 28 and 35 days. Experimental group rats long wear appliance all day and control group rats treated with nothing. And all the rats free diet and drinking water. The rats were sacrificed by cervical dislocation in the period of the experiment, to obtain the condylar tissue, HE staining and SP immunohistochemistry to detect the expression of ERK and integrin β1 staining results. Results: The surface of rat condylar cartilage showed a distinct layer of fibrous tissue in control group, the anterior portion of the condylar cartilage is thin. The condylar cartilage thickness of experimental groups at each time point increased significantly. 7, 14, 21 and 28 days, ERK and integrin β1 expression level of the observation group in rat tissue was significant higher than the control group (P<0.05). But there was no significant difference between the two groups of ERK and integrin β1 level at the 1, 3 and 35 day(P>0.05). At the 7~14 days in experimental group, ERK and integrin β1 peak, then decreased gradually. Conclusion: The increment activity of the posterior condylar cells changes under stress, and is closely related to the expression of ERK and integrin β1, which may play an important role in the signal transduction process.

摘要: 目的:建立SD大鼠下颌前伸动物模型,探讨ERK(细胞外调节蛋白激酶)与整合素β1的变化规律,分析其介导生长期大鼠下颌功能前伸后髁突骨形态学改变过程。方法:将42只4周龄SPF级健康雄性SD大鼠随机分为对照组及和实验组,每组3只,共14组。实验周期分别为1、3、7、14、21、28及35 d。实验组大鼠全天佩戴矫治器,对照组大鼠不做任何处理,所有大鼠自由饮食饮水。依照实验周期将各组大鼠脱颈椎后处死,取髁突组织,行HE染色及SP免疫组化法检测ERK及整合素β1染色结果。结果:对照组大鼠髁突软骨组织表面可见一层明显的纤维组织,髁突软骨前份最薄;各时间点实验组大鼠髁突软骨厚度明显增加。第7、14、21、28天时,观察组大鼠组织中ERK及整合素β1表达水平高于对照组,且差异存统计学意义(P<0.05),但第1、3及35天时两组ERK及整合素β1水平差异无统计学意义(P>0.05),且第7~14天时实验组ERK及整合素β1达到峰值,后逐渐降低。结论:髁突细胞增殖活性在应力作用下出现改变,且与ERK与整合素β1的表达密切相关,表明ERK与整合素β1在力学信号转导过程中也起到十分重要的作用。
