褪黑素对纯钛超声微弧氧化膜层体内植入骨结合的影响

Osteoblast Responses to Melatonin-coated Implants in vivo

摘要: 目的:将具有骨诱导活性的褪黑素引入纯钛超声微弧氧化膜层中,通过动物实验观察能否提高早期骨整合能力。方法:纯钛表面制备超声微弧氧化膜层(A组),再以硅烷为偶联剂(B组),进一步组装褪黑素(C组),植入新西兰大白兔的双侧下颌骨中2、4和6周,利用CBCT、扭力实验、甲苯胺蓝染色观察各组的种植体?鄄骨界面早期成骨情况。结果:纯钛超声微弧氧化膜层经硅烷偶联褪黑素制备的复合膜层表面保留微弧氧化的微孔形貌,增加了利于骨生成和生长的硅、碳和氮元素。添加褪黑素膜层骨结合能力最好,植入4周时的骨结合力达到峰值,其余2组植入6周时的骨结合力才达到峰值。添加褪黑素种植体骨界面新骨形成更早、骨量更多。结论:以硅烷为偶联剂在纯钛表面超声微弧氧化膜层表面组装褪黑素,获得的复合膜层种植体,能加速早期的骨组织的形成,具有更好的生物活性。

Abstract: Objective: This study aimed to evaluate the effect of biological activity and early bone binding capacity of Melatonin-coated titanium Implants with ultrasonic-micro-arc oxidation. Methods: Implants coated with ultrasonic-micro-arc oxidation-silane are regared as Group B, and further assembly of melatonin as group C. The control group A implants with only the ultrasonic-micro-arc oxidation. Implants were installed into the proximal metaphyseal area of each tibia of New Zealand white rabbits. Observation and comparison  of osteoblasts responses  were  carried on at 2 weeks, 4 weeks, and 6 weeks by CBCT examination, torsion test, and toluidine blue staining. Results: The implants treated with ultrasonic-micro-arc oxidation- silane, and the ones further assembly with melatonin kept the original pore morphology of micro-arc oxidation. The composite coating increased the Si, C and N chemical elements. It was beneficial to the growth of new bone. The osseointegration of implants that joined with melatonin was the best. The osseointegration process  appeared the highest at the 4th week. The other two groups need 6 weeks to reach the same level of group C. The implants treated with melatonin achieved the best result in promoting the osseointegration, also showed the best biocompatibility. Conclusion: Implant treated with ultrasonic-micro-arc oxidation- silane, and further assembly of melatonin can accelerate osteointegration.
