Nestin阳性细胞中DMP1过表达对小鼠软骨细胞分化的影响

Overexpression of DMP1 in Nestin Positive Cells Promotes Chondrocyte Differentiation in Mice

摘要: 目的:细胞外基质蛋白对软骨发育有重要调控作用。近期研究发现DMP1是软骨发育过程中特异性表达的基质蛋白,但其对软骨细胞分化的作用尚未清楚。Nestin阳性细胞是间充质干细胞中重要的一个亚群,我们通过在Nestin阳性细胞中过表达DMP1,研究DMP1对软骨细胞分化的影响。方法:以Nestin为启动子构建DMP1转基因小鼠模型,通过组织学切片染色观察小鼠关节软骨结构和细胞形态的变化,实时荧光定量PCR分析相关基因的表达差异。结果:DMP1转基因小鼠关节软骨层增厚,软骨细胞数量增加,Sox9、Col Ⅱ、Col X、Acan等软骨细胞分化相关基因表达上调。结论:DMP1能够促进小鼠Nestin阳性的软骨细胞分化,提示DMP1对软骨发育可能具有调控作用。

Abstract: Objective: To study whether the overexpressed Dentin Matrix Protein (dmp1 )in nestin positive cells was related to the promotion of chondrocyte differentiation. Methods: DMP1 transgene mice model was constructed by nestin promoter. Histological section and staining were used to observe the change of mice articular cartilage structure and cell morphology. Real-time quantitative PCR were performed to analyze the differences of related gene expression. Results: The thickness of DMP1 transgene mice articular cartilage increased, hypertrophic chondrocyte number increased and gene expression of Sox 9, Col Ⅱ, Col X, and Acan was up-regulated. Conclusion: DMP1 promotes nestin positive chondrocyte differentiation in mice, suggests DMP1 may regulate cartilage development.
