不同静压力下髁突软骨细胞MMP-13的表达变化研究

The Effect of the Static Pressure on Expression of MMP-13 in Condylar Chondrocytes

摘要: 目的:探讨在不同强度及不同时间的静压力作用下,髁突软骨细胞中MMP-13表达的变化及意义。方法:体外分离培养SD大鼠髁突软骨细胞,用甲苯胺蓝及Ⅱ型胶原免疫细胞化学染色进行鉴定;分别用0、50、100、150、200 kPa静压力进行2 h的加压处理。另外在200 kPa静压力下,分别对髁突软骨细胞进行0、1、2、4、8和12 h的加压,采用Western blot免疫印迹及实时荧光定量PCR,分析髁突软骨细胞在不同强度及不同时间的静压力加载后,MMP-13的mRNA和蛋白表达变化。结果:在50~200 kPa的静压力范围内,随着压力的增大,MMP-13蛋白及mRNA表达水平均呈下降趋势,实验组表达水平均降至对照组(0 kPa组)以下,且差异有统计学意义(P<0.05)。在200 kPa的静压力下,随着加压时间的延长,MMP-13表达呈先上升后下降的趋势,且4 h实验组达最高水平(P<0.05)。结论:髁突软骨细胞对压力微环境的改变具有较好的适应能力,压力的改变可能影响颞下颌关节的适应性改建。

Abstract: Objective: To investigate the effect of the static pressure on the mRNA and protein expression of MMP-13 in condylar chondrocytes. Methods: Condylar chondrocytes were dissected and cultured from the TMJ of 3-week-old SD rat in vitro. The chondrocytes of P3 were identified and were subjected to different static pressure of 0, 50, 100, 150 and 200 kPa respectively for 2 h, while the monolayer of condylar chondrocytes were subjected to static pressure of 200 kPa for different time (0, 1, 2, 4, 8 and 12 h). Expression of MMP-13 was examined by Western blot and quantitative real-time PCR. Results: The static pressure with a magnitude from 50 to 200 kPa can reduce the expression of MMP-13 of condylar chondrocytes, and has a significant difference (P<0.05). Under static pressure of 200 kPa, with the compression time extended, the expression of MMP-13 first increased and then decreased, and up to the highest level at 4 h(P<0.05). Conclusion: The results indicated TMJ condylar chondrocytes has a certain adaptability to static compression.
