The expression and clinical significance of PTK7 in oral squamous cell carcinoma

摘要:

目的:通过在口腔鳞状细胞癌(oral squamous cell carcinoma, OSCC)组织样本中研究酪氨酸蛋白激酶7(protein tyrosine kinase 7, PTK7)的表达,分析PTK7蛋白的表达在OSCC中的作用及对患者预后的影响。方法:选取OSCC组织样本总共113例,采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结(streptavidin perosidase, SP)法检测PTK7蛋白的表达,结合OSCC发展、预后等临床相关因素进行卡方检验及Kaplan-Meier生存分析等统计学分析。结果:113例OSCC组织标本中有70例PTK7蛋白表达阳性,在癌旁正常鳞状上皮组织中,PTK7蛋白表达为阴性。进一步的分析结果表明,PTK7的表达与OSCC患者的相关影响因素(年龄、性别、分化程度、临床分期、肿瘤大小及局部浸润范围、淋巴结转移等)相比较,差异无统计学意义(P≥0.05);Kaplan-Meier生存分析结果显示,PTK7蛋白表达阴性组的总生存率显著高于阳性组(P<0.05)。结论:PTK7的检测有助于综合判断OSCC的临床预后,提示PTK7作为OSCC的预后指标值得深入研究。

Abstract:

Objective: To evaluate the expression of protein tyrosine kinase 7 (PTK7) in oral squamous cell carcinoma (OSCC), and to analyze the role of PTK7 protein expression in OSCC and its impact on the prognosis of patients. Methods: We collected 113 specimens of OSCC, and the PTK7 protein expression was determined by streptavidin perosidase (SP) method. The chi-square test and Kaplan-Meier analysis were used for the analysis of the expression of PTK7 protein and the OSCC clinical related factors. Results: In the 113 OSCC specimens, PTK7 protein expressed positive was found in 70 cases, PTK7 protein expressed negative was found in the para-cancer area. The PTK7 expression intensity was not significantly associated with the patients' age, gender,differentiation degree, clinical stage, tumor size, local infiltration extent and lymph node metastasis (P≥0.05). Kaplan-Meier survival analysis showed that the survival rate of PTK7 negative patients was significantly higher than that of positive patients (P<0.05). Conclusion: These findings suggested that the detection of PTK7 protein may aid in the comprehensive assessment of OSCC outcome.Further research for direct measurement are needed.
