miR-378a在口腔鳞状细胞癌中的表达及意义

The expression and significance of miR-378a in oral squamous cell carcinoma

摘要:

目的:探讨口腔鳞状细胞癌(oral squamous cell carcinoma, OSCC)组织中miR-378a的表达,以及其与患者预后的相关性。方法:选取我院2015年3月—2018年3月择期行手术治疗的91例OSCC患者,实时定量聚合酶链反应(real-time quantitative polymerase chain reaction,RT-qPCR)检测OSCC和癌旁组织中miR-378a的表达情况。于术后第1天开始对患者进行随访,随访截至2021年3月31日,记录患者总生存时间和死亡情况。结果:OSCC组织中miR-378a的相对表达量低于其在癌旁组织中(P<0.001);miR-378a的相对表达量在肿瘤不同TNM分期、分化程度和淋巴结转移方面,差异有统计学意义(P<0.05);Kaplan-Meier分析结果显示,高表达组患者平均生存时间为32.29个月,3年生存率为73.81%,高于低表达组的24.80个月和30.61%,差异有统计学意义(P<0.001);miR-378a的表达、TNM分期、分化程度及淋巴结转移是影响患者生存时间的风险因素(P<0.05)。结论:miR-378a在OSCC组织中的表达量低于其在正常组织中的表达量,且其与肿瘤TNM分期、分化程度及淋巴结转移相关,是影响患者预后的风险因素。

Abstract:

Objective: To investigate the expression of miR-378a in oral squamous cell carcinoma (OSCC) tissues and its correlation with the prognosis of patients. Methods: Subject recruitment consisted of 91 patients with OSCC, surgical procedures were carried out from March 2015 to March 2018. Real-time quantitative polymerase chain reaction (RT-qPCR) was used to detect the expressions of miR-378a in OSCC and adjacent tissues. The patients were followed up on the first day after operation, and the deadline for follow-up was March 31, 2021. With death or expiration of follow-up as the end event, the death and overall survival time during the follow-up period were recorded. Results: The relative expression level of miR-378a in OSCC tissues was lower than that in the adjacent tissues (P<0.001). The differences of the relative expression levels of miR-378a in different TNM stage, degree of differentiation and lymph node metastasis were statistically significant (P<0.05). Kaplan-Meier analysis showed that the average survival time of patients in the high expression group was 32.29 months, and the 3-year survival rate was 73.81%, which was higher than 24.80 months and 30.61% in the low expression group (P<0.001). The expression of miR-378a, TNM stage, degree of differentiation and lymph node metastasis were risk factors that affect the survival time of patients (P<0.05). Conclusion: The present study revealed that the expression level of miR-378a in the tissues of OSCC is weaker than that in the normal tissues. The expression intensity is related to TNM stage, degree of differentiation and lymph node metastasis. It is a risk factor that affects the prognosis of patients.
