浓缩生长因子对人上颌窦黏膜细胞增殖迁移

摘要: 目的:探讨浓缩生长因子提取液(concentrated growth factors extract, CGFe)对上颌窦黏膜细胞增殖、迁移的影响。方法:体外培养人上颌窦黏膜细胞,实验组采用含CGFe的完全培养基,对照组采用不含CGFe的完全培养基培养。通过CCK-8法(cell counting kit-8, CCK-8)、细胞周期检测法评估细胞增殖速率变化,划痕实验评估细胞迁移速率,免疫荧光、实时荧光定量PCR法(real-time PCR,RT-PCR),检测上颌窦黏膜细胞表达增殖相关的基因的变化。结果:实验组的细胞增殖、迁移速率及增殖相关标记物的表达均高于对照组。结论:浓缩生长因子提取液能有效促进上颌窦黏膜细胞的增殖、迁移。

Abstract: Objective: To study the effects of concentrated growth factors extract (CGFe) on the proliferation and migration of maxillary sinus mucosa cells. Methods: Human maxillary sinus mucosa cells were cultured in vitro, and the experimental group was treated with containing CGFe, while the control group was treated with without CGFe. The variation of cell proliferation was detected by cell counting kit-8 (cell counting kit-8, CCK-8) and cell cycle assay. Besides, the scratch experiment was used to evaluate thecell migration rate, and immunofluorescence and real-time PCR (RT-PCR) were applied to detect the variation of gene expression related to proliferation in maxillary sinus mucosa cells. Results: The cell proliferation, migration rate and proliferation related markers in the experimental group were higher than those in the control group. Conclusion: Concentrated growth factors extract was effective in promoting proliferation and migration of maxillary sinus mucosa cells.
