Hydrogel Biomimic Microenvironments of Stem Cell for Bone Tissue Engineering

摘要:

 间充质干细胞(mesenchymal stem cells,MSCs)具有自我复制和多向分化潜能的特点,水凝胶被广泛用于负载MSCs通过细胞治疗和组织工程修复各类口腔疾病,同时被用于仿生构建MSCs微环境,作为研究干细胞分化调控机制的工具,促进定向调控干细胞分化培养体系的发展。MSCs生存在一个复杂的微环境中,其生命活动受到微环境信号的调控,通过构建水凝胶载体模仿MSCs微环境,根据不同应用可以在一定程度上调控MSCs的分化潜能。本综述概括了目前关于利用水凝胶仿生设计MSCs微环境的研究进展,总结了用于负载MSCs水凝胶的材料性能、材料设计及其在骨组织修复中的应用。

Abstract:

 Mesenchymal stem cells are characterized by their dual ability:  self-renewal and differentiation. Hydrogels are wildely used as carriers for MSCs in cell therapy and tissue engineering in regenerative medicine. Meanwhile, such synthetic hydrogels can biomimicking microenvironments of MSCs, becoming powerful tools for elucidating MSCs regulatory mechanisms and accelerate the development of advanced culture systems. MSCs live in a complex "niche", and their fate was regulated by the "niche" signals. This article reviews the biomaterials and design of hydrogels used for biomimicking microenvironments, highlighting the application in bone tissue engineering.
