SOX18通过上调KLF4促进血管瘤内皮细胞增殖并抑制其凋亡

SOX18 Promotes Endothelial Cells of Hemangioma Proliferation and Suppresses Apoptosis Through Targeting KLF4

摘要: 目的:构建表达SOX18的真核表达载体,并合成靶向于SOX18的siRNA,探索SOX18调控血管瘤内皮细胞(hemangioma-derived endothelial cell, HemECs)增殖的机制。方法:检测SOX18及相关基因在临床样本中的表达。构建SOX18的真核表达载体,通过转染该真核表达载体以及靶向于SOX18的siRNA,实现过表达和敲减SOX18。以细胞计数的方法检测SOX18对血管内皮细胞增殖的影响;流式细胞术检测超表达和敲减SOX18后对细胞凋亡的影响;qRT-PCR检测过表达和敲减SOX18后对相关基因的调控作用。结果:SOX18在血管瘤组织总的表达显著高于正常组织。成功构建了过表达SOX18的真核表达载体pCMV-HA-SOX18,并验证其能够在HemECs细胞内过表达。此外,结合siRNA敲减实验,证实SOX18能够促进HemECs增殖并抑制其凋亡。qRT-PCR检测和Western Blot检测证明SOX18能够上调KLF4和VEGF-C的表达。结论:SOX18能够通过促进KLF4和VEGF-C的表达促进HemECs增殖。

Abstract: Objective: To construct the recombinant plasmid containing SOX18 gene and investigate the effect of the protein on vascular endothelial cells from the source of hemangiomas. To explore the mechanism of SOX18 in regulating the proliferation of vascular endothelial cells. Methods: qRT-PCR was used to detect the SOX18 expression levels in hemangiomas tissues and adjacent tissues. The SOX18-overexpressing recombinant plasmid pCMV-HA-SOX18 or specific siRNA targeting SOX18 were transfected into vascular endothelial cells. Cell counting assay was performed to evaluate the cell proliferation. The cell apoptosis was detected by flow cytometry method. qRT-PCR was used to determine the expression of the cell proliferation related genes. Results: The SOX18 expression was significantly increased in hemangiomas tissues. Overexpression of SOX18 promoted cell proliferation and inhibited apoptosis in vascular endothelial cell. qRT-PCR and Western blot results suggest that SOX18 could increase the expressions of KLF4 and VEGF-C. Conclusion: SOX18 could promote KLF4 and VEGF-C expression to affect cell proliferation.
