Expression and effect of CYP3A5 gene in oral squamous cell carcinoma

摘要:

目的:探究CYP3A5在口腔鳞状细胞癌(oral squamous cell carcinoma, OSCC)组织及细胞中的表达及其对OSCC细胞生物学行为能力的影响。方法:采用实时荧光定量聚合酶链反应(real-time quantitative polymerase chain reaction, RT-qPCR)法和蛋白质免疫印迹(Western blotting)法分别检测OSCC组织及细胞中CYP3A5基因和蛋白的表达,通过CYP3A5 siRNA敲除降低CYP3A5表达水平,观察CYP3A5基因对OSCC细胞增殖、侵袭、迁移等能力的影响。通过免疫组织化学(immunohistochemistry,IHC)法检测OSCC及癌旁组织中CYP3A5的表达水平。结果:与癌旁组织相比,OSCC组织中的CYP3A5呈明显低表达(P<0.05);与正常口腔上皮细胞系HOK相比,OSCC细胞系SCC-9、SCC-25、CAL-27中CYP3A5的表达水平明显偏低(P<0.05)。抑制CYP3A5基因表达能明显促进SCC-9、SCC-25、CAL-27的增殖、侵袭和迁移能力(P<0.05)。免疫组织化学分析结果显示,CYP3A5在OSCC癌组织中呈显著低水平。结论:CYP3A5在OSCC组织和细胞中呈明显低表达,对于促进OSCC的增殖、侵袭和迁移具有重要意义。

Abstract:

Objective: To investigate the expression level of CYP3A5 in oral squamous cell carcinoma (OSCC) tissues and cells and its effects on the biological behavior of OSCC. Methods: Real-time quantitative polymerase chain reaction (RT-qPCR) and Western blotting were used to detect the genetic and protein expression of CYP3A5 in OSCC tissues and cells respectively, CYP3A5 siRNA was used to decrease the expression level of CYP3A5, and the effects of CYP3A5 gene on the proliferation, invasion and migration of OSCC cells were observed. The expression levels of CYP3A5 in OSCC and adjacent tissues were detected by immunohistochemistry(IHC). Results: Compared with adjacent cancer tissues, the genetic expression level of CYP3A5 was significantly lower in OSCC tissues(P<0.05). Compared with normal oral epithelial cell line HOK, the expression levels of CYP3A5 gene in OSCC cell lines SCC-9, SCC-25, and CAL-27 were significantly lower (P<0.05). Inhibiting the expression level of CYP3A5 gene significantly promoted the proliferation, invasion and migration capability of SCC-9, SCC-25, and CAL-27 cell lines(P<0.05). Immunohistochemical analysis showed that CYP3A5 demonstrated weaker signal in OSCC(P<0.05). Conclusion: The expression level of CYP3A5 was significantly lower in OSCC tissues and cells, which was of great significance for promoting the proliferation, invasion and migration of OSCC.
