闭锁小带蛋白-1基因在口腔疣状癌和口腔鳞癌中的转录水平分析

Identification of ZO-1 in Oral Verrucous Carcinoma and Oral Squamous Cell Carcinoma

摘要: 目的:研究闭锁小带蛋白-1(zonula occluden -1,ZO-1)在口腔疣状癌和口腔鳞癌中的表达,分析ZO-1相关的上皮间充质转化过程参与口腔癌发生发展的情况。方法:取20例口腔鳞癌及其癌旁组织,10例口腔疣状癌组织及10例正常口腔黏膜组织标本,荧光实时定量Real-time PCR方法检测ZO-1基因mRNA在不同组织中的表达。结果:口腔疣状癌与鳞癌组织中ZO-1基因mRNA的表达低于正常口腔黏膜上皮组织与癌旁组织(P<0.05);口腔疣状癌中ZO-1基因mRNA的表达高于鳞癌(P<0.05)。结论:ZO-1作为上皮间充质转化过程中的关键蛋白,其在口腔疣状癌与鳞癌中表达水平的差异,表明口腔鳞癌与口腔疣状癌中存在不同程度的上皮间充质转化现象。

Abstract: Objective: This study was purposed to analysis the transcriptional expression of Zonula Occluden -1 (ZO-1) in oral verrucous carcinoma(OVC) and oral squamous cell carcinoma(OSCC). Methods: Pathological tissue samples were obtained from patients of OSCC(n=20), OVC(n=10), as well as matched OSCC pericarcinomatous tissue(n=20) and normal oral mucosa (n=10). The transcriptional expression of ZO-1 was detected by Real-time PCR method. Results: The transcriptional expressions of ZO-1 in OSCC and OVC were lower than their matched pericarcinomatous tissue and normal oral mucosa tissue (P<0.001). The ZO-1 transcriptional level in OSCC was significantly lower than that in OVC (P<0.05). Conclusion: There might be different epithelial-mesenchymal transition process among OVC and OSCC.
