HIF-1α Promotes Migration and Proliferation of Oral Squamous Cell Carcinoma Cell by Increasing the Expression Level of MiR-210-3p

摘要:

目的:研究低氧诱导因子-1α(hypoxia-inducible factor 1α,HIF-1α)是否通过改变miR-210-3p的表达水平影响口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)的进展。方法:低氧培养CAL-27细胞并检测细胞内HIF-1α、miR-210-3p的表达水平;重组HIF-1α刺激CAL-27细胞后检测miR-210-3p的表达水平是否发生变化;转染短发夹RNA(short hairpin RNA,shRNA)改变CAL-27细胞内miR-210-3p的表达水平后,用划痕实验和CCK-8实验来观察其对CAL-27细胞迁移、增殖能力的影响。结果:低氧导致CAL-27细胞内HIF-1α及miR-210-3p表达量增高;重组HIF-1α处理后的CAL-27细胞内miR-210-3p表达量升高;过表达miR-210-3p会促进CAL-27细胞的增殖及迁移能力,反之细胞的增殖和迁移能力则受到抑制。结论:MiR-210-3p在OSCC细胞增殖和迁移过程中起到重要作用,HIF-1α通过影响miR-210-3p表达来参与肿瘤进程。

Abstract:

Objective: To study whether hypoxia-inducible factor 1α( HIF-1α) affects the progress of oral squamous cell carcinoma(OSCC) by changing the expression level of miR-210-3p. Methods: OSCC CAL-27 cell line was cultured with hypoxia and the expression level of HIF-1α and miR-210-3p was detected. After stimulated by recombinant HIF-1α, we detected the expression level of miR-210-3p in CAL-27 cells. And the effects of short hairpin RNA (shRNA) on the migration and proliferation of CAL-27 cells were observed by Scratch and CCK-8 assay after shRNA transfer changed the expression level of miR-210-3p in CAL-27 cells. Results: Hypoxia condition increased the expression level of HIF-1α and miR-210-3p in CAL-27 cells. The expression intensity of miR-210-3p increased in CAL-27 cells treated with recombinant HIF-1α. Overexpression of miR-210-3p promoted the proliferation and migration of CAL-27 cells, while the proliferation and migration of CAL-27 cells were inhibited by reproduction. Conclusion: MiR-210-3p plays an important role in the proliferation and migration of OSCC cells, and HIF-1α participates in the tumor process by affecting the expression level of miR-210-3p.
