Cr 3+与EGCG共同作用对牙龈成纤维细胞67LR和EGFR表达的影响

Complexation of Epigallocatechin-3-gallate with Cr3+ on the Expression of 67LR and EGFR on Gingival Fibroblast Cells

摘要: 目的: 分析表没食子儿茶素没食子酸酯(EGCG)与Cr3+共同作用,对牙龈成纤维细胞(HGF)的EGCG受体67LR和表皮生长因子受体(EGFR)的影响。方法:EGCG与Cr3+共同作用牙龈成纤维细胞24 h后,用Real-time PCR法检测mRNA表达,运用Western印迹法分析蛋白表达。结果:EGCG能抑制HGF细胞表面受体67LR mRNA以及蛋白水平的表达,加入Cr3+后减弱了EGCG对细胞67LR mRNA和蛋白的影响。EGCG作用24h能降低EGFR mRNA和蛋白的表达,Cr3+(<200 μmol/L)会减弱EGCG抑制EGFR的作用。结论:EGCG能降低HGF细胞67LR和EGFR的表达。加入 Cr3+后对细胞67LR无明显影响,但降低细胞EGFR的表达。

Abstract: Objective: The purpose of this study was to examine the complexation effects of epigallocatechin-3-gallate (EGCG) with Cr 3+ on the expression of 67LR and EGFR on human gingival fibroblast cells. Methods: Real-time PCR was adopted to study the expression of 67LR, EGFR at mRNA levels and Western blot on protein levels. Results: EGCG suppressed the expression of 67LR, EGFR mRNA and proteins. The co-treatment of Cr 3+ and EGCG decreased EGCG-induced inhibition effect on 67LR expression. Cr 3+ in low concentration (lesser than 200 μmol/L) could decrease EGCG-induced inhibition effect on EGFR expression. Conclusion: EGCG inhibits the expression of 67LR and EGFR. EGCG mixed with Cr 3+ could reduce EGFR eexpression and has no significant effects on 67LR expression.
