淫羊藿苷对小鼠骨髓腔中成骨和成脂分化作用的研究

Icariin Regulates the Osteogenesis and Adipogenesis in the Bone marrow of Osteoporotic Mice

摘要: 目的:通过建立小鼠骨质疏松模型,研究口服淫羊藿苷(Icariin,ICA)对小鼠骨髓腔中成骨分化和成脂分化中的作用。方法:将36只小鼠随机分为3组,分别为假手术组(SHAM组)、卵巢切除组(OVX组)和卵巢切除并灌服淫羊藿苷组(ICA组)。ICA组按每kg体质量25 mg的剂量给予淫羊藿苷每日灌服,SHAM组和OVX组每日给予等体积0.9%氯化钠溶液灌服,于术后9周收样。取左侧股骨和胫骨进行切片染色,提取右侧骨组织中mRNA和蛋白,分析成骨和成脂基因和蛋白的变化。结果:①术后5周,OVX组体重显著高于SHAM组和ICA组(P<0.05)。②HE染色显示,与OVX组相比,ICA组骨髓腔中骨小梁生成增加了50%,脂肪细胞生成减少了70%。③q-PCR和Western blot 结果分析显示,与OVX组相比,ICA组骨组织中成骨相关的基因和蛋白表达量显著升高,而成脂相关的基因和蛋白表达量显著降低。结论:口服淫羊藿苷可以通过促进骨吸收并抑制骨髓腔中脂肪细胞的形成,从而对预防和治疗骨质疏松具有重要的意义。

Abstract: Objective: To study the role of Icariin (ICA) on the adipogenesis and osteogenesis in the bone marrow by using the murine osteoporosis model. Methods: 36 mice were divided into 3 groups: Sham-operated group (SHAM), ovariectomized group (OVX) and Icariin administered with ovariectomized group (ICA). Mice in the ICA group were treated with ICA by oral administration; the other two groups were treated with the same amount of normal saline. Mice samples were harvested in 9 weeks after operation. Left femurs and tibias were used for HE staining. Right femurs and tibias were used to harvest tissue protein and mRNA to detect the expressions changes of genes and proteins. Results: OVX mice had more body weight than other two groups 5 weeks after the operation (P<0.05). HE staining showed: mice in ICA group had significantly increased trabecular bone formation and decreased adipocytes in the bone marrow compared with OVX group (P<0.05). The results of qPCR and Western blot showed: compared with OVX group, mice in the ICA group have more expression of osteogenic-related genes and proteins accompanied by the decreased expression of adipogenic-related genes and proteins. Conclusion: Oral administration of ICA can improve the conditions of osteoporosis by promoting osteogenesis and inhibiting adipogenesis in the bone marrow.
