不同浓度淫羊藿浸泡种植体对骨结合影响的实验研究

The Study of Osseointegration Responses to Icariin-coated Implants

摘要: 目的:通过比较种植体表面淫羊藿苷载药浓度对骨结合的影响,探究种植体植入时,淫羊藿苷加载的最适宜浓度。方法:将事先制备好的36枚种植体随机分成6组,分别浸泡于对照组及用无水乙醇配制的20、40、60、80、100 mg/L的淫羊藿苷溶液中,随机植入18只家兔股骨内。于第3周末进行荧光四环素(50 mg/Kg)肌肉注射,4周时处死实验动物。通过大体、硬组织切片及旋出扭矩测试,进行比较分析。结果:用60~80 mg/L淫羊藿苷溶液处理过的种植体,其成骨量及骨结合水平优于其他浓度组。结论:淫羊藿苷能促进成骨,在一定浓度内其成骨作用与药物浓度呈正相关。而高浓度的淫羊藿苷则能抑制成骨。

Abstract: Objective: This study aimed to find out the appropriate concentration of icariin-coated implants to maximize improvement of osteointegration. Methods: 36 implants were randomly allocated into 6 groups: implants of control group (n=6)  were coated by anhydrous ethanol; implants of experiment group 1 were coated by icariin solution of 20 mg/L, implants of group 2 were coated by icariin solution of 40 mg/L, in group 3 was 60 mg/L, in group 4 was 80 mg/L, and group 5 was 100 mg/L (n=6 respectively). Implants were inserted into 18 New Zealand white rabbit femurs  stochastically. After 3 weeks of healing, the experimental subjects were injected with tetracyline fluorescence (50 mg/kg), and  sacrificed at 1 week later.  Gross observation, hard tissue slicing and conventional removal torque(RTQ) evaluation  were performed for group comparing. Results: Group 3  and 4, icariin solution 60-80 mg/L coated implants showed the greatest new bone formation and osseointegration level. Conclusion: Icariin can be used to hasten  the osseointegration between implant and bone. Within certain extent, the osseointegration is positively associated with the concentration of icariin, while, high level icariin may  be negative to bone formation.
